A flame atomic spectrophotometer is an instrument based on the principles of atomic absorption spectroscopy to determine the content of a specific element in a sample. The instrument atomizes the sample solution and introduces it into a high-temperature flame to atomize the elements to be measured, and subsequently measures the absorption of these ground state atoms to the characteristic wavelength light, enabling quantitative analysis of elemental concentrations. Its detection scope usually covers a variety of metals and some semi-metallic elements, and is widely used in environmental monitoring, food safety, materials science, and industrial quality control.
Principle
The working principle of the flame atomic spectrophotometer is based on atomic absorption spectroscopy. When the sample solution forms an aerosol through an atomizer and enters the flame with the carrier gas, the energy provided by the high-temperature flame dissociates the element to be measured into ground state atoms. These ground-state atoms selectively absorb specific wavelengths of resonant radiated light emitted by hollow cathode lamps, resulting in a decrease in light intensity. The relationship between the absorbance value and the atomic concentration of the element to be measured in the sample follows Lambert-Beale's law, and its relationship can be expressed as:

where A is the absorbance, I₀ is the intensity of the incident light, I is the intensity of the transmitted light, k is the absorbance coefficient, c is the concentration of the element to be measured, and l is the length of the absorbed optical path. Measurement method
The routine measurement process includes sample preparation, instrument calibration, measurement and data analysis steps. Samples are usually pre-treated with digestion, filtration, and dilution to transform into a homogeneous solution. The instrument establishes a standard operating curve for absorbance versus concentration using a range of standard solutions at known concentrations. When measuring, the sample solution is drawn into the atomization system, mixed with gas and auxiliary gas in a mixing chamber and then entered the burner, where it is atomized in the flame. The spectroscopic system separates the light emitted by the hollow cathode lamp into characteristic wavelengths, and the detector measures the change in light intensity and converts it into absorbance values. By comparing the standard curves, the concentration of elements in the sample can be calculated. To reduce interference, standard addition or background correction techniques are often used.
Influencing factors
The accuracy and repeatability of measurement results are affected by a variety of factors. The flame conditions include the type and ratio of gas and auxiliary gas, such as acetylene-air or acetylene-nitrous oxide combination, and its temperature and reducing properties directly affect the atomization efficiency. The performance of the atomization system determines the stability of sample introduction and the atomization particle size. The wavelength accuracy and spectral bandwidth of the optical system affect the resolution ability. Chemical interference can result from ionization effects or compound formation caused by other components in the sample matrix, which can be mitigated by adding release agents or protective agents. Physical disturbances include changes in solution viscosity, surface tension, and transfer rate, which can be mitigated by keeping the standard solution matched to the sample matrix. The stability of instrument parameters such as lamp current, slit width and detector response should also be checked regularly.
