Light-sheet microscope

Light sheet microscopy uses a thin sheet of laser light to illuminate the sample, exciting fluorescence only at the focal plane, while a perpendicular camera collects the signal. This method reduces photodamage to the sample and minimizes background interference, making it suitable for observing the three-dimensional structure and dynamic processes of living biological samples.
Selection
Select the laser wavelength with matching penetration power based on sample thickness, choose the scanning system according to imaging speed requirements, and determine the light sheet thickness based on sample transparency. Consider the compatibility between camera sensitivity and objective numerical aperture to ensure the system is compatible with existing fluorescence labeling methods.
